Mechanism of Action
NiaXtend® extended-release niacin reduces cholesterol levels through multiple mechanisms that target lipid metabolism. It inhibits hormone-sensitive lipase in adipose tissue, reducing the release of free fatty acids into the bloodstream. This leads to decreased triglyceride production in the liver, which in turn reduces the synthesis of very low-density lipoproteins (VLDL) and low-density lipoproteins (LDL). Niacin also increases lipoprotein lipase activity, enhancing the clearance of triglycerides from the blood.
Additionally, it decreases the hepatic production of ApoB, a key protein for VLDL and LDL formation, further lowering LDL levels. Niacin boosts high-density lipoprotein (HDL) levels by slowing HDL clearance, promoting reverse cholesterol transport. It also uniquely lowers lipoprotein(a), a genetic risk factor for cardiovascular disease. Overall, niacin improves lipid profiles by reducing LDL, triglycerides, and Lp(a), while increasing HDL cholesterol.

Formulating with NiaXtend®
NiaXtend® is a proprietary, patented, vegetable-based extended-release wax matrix tablet available in 500 mg and 750 mg doses. They will dissolve in a slow and controlled manner releasing a minimum of 90% niacin by 7 hours of digestion. This helps to ensure optimal niacin absorption without the typical side effects caused by immediate release niacin. The 500 mg niacin-containing tablet weighs approximately 665 mg, while the 750 mg niacin-containing tablet weighs approximately 1,000 mg. The tablets are slightly mottled, off white, and round (500 mg) and oval (750 mg). The tablets have 5 years of shelf life.
Regulatory
NiaXtend® has been shown to be generally well tolerated, with lower incidences of flushing and gastrointestinal side effects (<10%) compared to immediate-release niacin. NiaXtend® is self-affirmed GRAS.
